LiSpark™ SuFi PCR Master Mixes

LiSparkTM Ultra-Fidelity DNA Polymerase is a superior enzyme in this category for robust PCR with extreme fidelity, with an error rate 108-fold lower than that of Taq polymerase. LiSparkTM Max Super-Fidelity DNA Polymerase possesses 5´→ 3´polymerase activity, 3´→5´exonuclease activity and will generate blunt-ended products. This product also capable of amplifying long fragments such as 20kb genomic DNA and 40kb λDNA. The recommended applications include high yield PCR, GC-rich PCR, site-directed mutagenesis and high throughput PCR, etc.

PCR reaction setup for amplification:
• 2× LiSpark Buffer: 25 µl
• dNTP Mix (10 mM each ): 25 µl
• F1: 2 µl
• R1 : 2 µl
• Human gDNA : 1 
• ddH2O: 18 

PCR cycling for amplification:
1. 95°C: 3 min
2. 35 x
    1. Denaturation: 15 s at 95°C
    2. Annealing: 15 s at 63°C
    3. Extension: 30 sec/kb at 72°C
3. Final extension: 5 min at 72°C

From the above results, it can be concluded that LiSparkTM is suitable for the amplification of a variety of difficult templates. Under high GC, low template concentration and high inhibitor conditions, LiSparkTM DNA polymerase invariantly performed better than other polymerases. Although direct amplification of lysates is common practice for shorter fragments, amplification of longer fragments is more consistent with LiSparkTM DNA polymerase.
