Description:   This recombinant human adenovirus type 5 expresses a codon-optimized Cas9 nuclease fusion with GFP as reporter under CMV promote and can be used along with guided RNA for genome-editing.The Type II prokaryotic CRISPR/Cas system is the new class of tools for targeted genome engineering. Cas9 (CRISPR associated protein 9) is an RNA-guided DNA nuclease enzyme associated with Streptococcus pyogenes' CRISPR immunity system. The cas9 nucleases derived from clustered regularly interspaced short palindromic repeats (CRISPR)-cas systems uses small RNAs as guides (gRNA) to cleave DNA in a sequence-specific manner. With its ease in designing guide sequences to target specific genomic loci, the CRISPR/Cas system is a much simpler, faster, and robust alternative to TALEN and Zinc finger nuclease platforms.

Background:   Cas9 is a RNA-guided DNA endonuclease enzyme associated with the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) adaptive immunity system in bacteria and genetic engineering. Staphylococcus aureus derived Cas9 has a number of properties that make it advantageous for genome editing, including its small size, high efficiency, nickase activity, and apparent specificity. SaCas9 principally recognizes an NNGRRT protospacer adjacent motif (PAM) and cleaves target DNA at rates comparable to SpCas9 (Cas9 derived from S. pyogenes) with an ideal spacer lengths of 24 to 20 nucleotides. In comparison to SpCas9, SaCas9 is smaller (1368 AA versus 1053 AA) and generates indels at a comparable rate when directed against target DNA with the mutually permissive NGGRRT PAM. This makes it the preferred enzyme for all-in-one delivery of Cas9 and multiple gRNA expression cassettes with Adeno-associated vectors.
