Background

The protein encoded by this gene is a ubiquitin-like protein that is conjugated to intracellular target proteins upon activation by interferon-alpha and interferon-beta. Several functions have been ascribed to the encoded protein, including chemotactic activity towards neutrophils, direction of ligated target proteins to intermediate filaments, cell-to-cell signaling, and antiviral activity during viral infections. While conjugates of this protein have been found to be noncovalently attached to intermediate filaments, this protein is sometimes secreted.

ISG15 antibodyWestern blotting analysis using anti-ISG15 antibody (Cat#62197). Total cell lysates (30 μg) from various cell lines were loaded and separated by SDS-PAGE. The blot was incubated with anti-ISG15 antibody (Cat#62197, 1:5,000) and HRP-conjugated goat anti-rabbit secondary antibody (Cat#201, 1:20,000) respectively. Image was developed using FeQ™ ECL Substrate Kit (Cat#226).

ISG15 expression in wild-type (WT) and ISG15 shRNA knockdown (KD) HepG2 cells with 20 μg of total cell lysates. β-Tubulin serves as a loading control. Immunocytochemical staining of HepG2 cells with anti-ISG15 antibody (Cat#62197, 1:1,000). Nuclei were stained blue with DAPI; ISG15 was stained magenta with Alexa Fluor® 647. Images were taken using Leica stellaris 5. Protein abundance based on laser Intensity and smart gain: Medium. Scale bar: 20 μm.
