Background

The protein encoded by the gene ADSL belongs to the lyase 1 family. It is an essential enzyme involved in purine metabolism, and catalyzes two non-sequential reactions in the de novo purine biosynthetic pathway: the conversion of succinylaminoimidazole carboxamide ribotide (SAICAR) to aminoimidazole carboxamide ribotide (AICAR) and the conversion of adenylosuccinate (S-AMP) to adenosine monophosphate (AMP). Mutations in this gene are associated with adenylosuccinase deficiency (ADSLD), a disorder marked with psychomotor retardation, epilepsy or autistic features. Alternatively spliced transcript variants have been found for this gene.

ADSL antibodyWestern blotting analysis using anti-adenylosuccinate lyase antibody (Cat#61256). Total cell lysates (30 μg) from various cell lines were loaded and separated by SDS-PAGE. The blot was incubated with anti-adenylosuccinate lyase antibody (Cat#61256, 1:5,000) and HRP-conjugated goat anti-mouse secondary antibody (Cat#101, 1:20,000) respectively. Image was developed using FeQ™ ECL Substrate Kit (Cat#226).
Western blotting analysis using anti-adenylosuccinate lyase antibody (Cat#61256). Adenylosuccinate lyase expression in wild type (WT) and adenylosuccinate lyase (ADSL) knockdown (KD) HSHC cells with 20 μg of total cell lysates. Hsp90 α serves as a loading control. The blot was incubated with anti-adenylosuccinate lyase antibody (Cat#61256, 1:5,000) and HRP-conjugated goat anti-mouse secondary antibody (Cat#101, 1:20,000) respectively. Image was developed using FeQ™ ECL Substrate Kit (Cat#226).
Flow cytometric analysis of Adenylosuccinate lyase expression in HeLa cells using anti-Adenylosuccinate lyase antibody (Cat# 61256, 1:2,000). Green, isotype control; red, Adenylosuccinate lyase.
